Introduction to Carbohydrate antigen 125 (CA125)

Carbohydrate antigen 125 (CA125) is a glycoprotein mainly composed of O bonds, and is encoded by the MUC16 gene. CA125 is also a discontinuous epitope of the tandem repeat domain on mucin 16 (muc16). Under natural conditions, it is a very large complex (possibly as much as 4 million Da, with core CA125 subunits exceeding 200,000 Da). CA125 undergoes autoproteolysis due to its intrinsic endogenous protease activity. The ectodomain of CA125 is hydrolyzed and released into the extracellular matrix, where it enters the circulatory system. CA125 is phosphorylated prior to release and dephosphorylated upon release. The release or secretion of CA125 is directly related to the epidermal growth factor receptor (EGFR) signaling pathway. Studies have revealed that CA125 has a significant effect on the migration of ovarian cancer cells. In addition, the presence of CA125 can inhibit the function of natural killer (NK) cells.

Reference Range

CA125 is the most common serum indicator for the diagnosis of ovarian cancer, and the higher the degree of inflammation, the higher level of CA125. Serum CA125 levels are applicable for diagnosis and treatment monitoring in most High-grade serous ovarian cancers. Changes in CA125 level can also be used for screening ovarian cancers.
